Heterologous production of equol by lactic acid bacteria strains in culture medium and food

The isoflavones daidzin and genistin, present in soybeans, can be transformed by the intestinal microbiota into equol 5-hydroxy-equol, compounds with enhanced availability bioactivity, although these are only produced a fraction of population. Hence, there is an interest production compounds, although, to date, few bacteria biotechnological applicability food have been found able produce equol. In order obtain lactic acid equol, daidzein reductase (dzr), dihydrodaidzein (ddr), tetrahydrodaidzein (tdr) racemase (ifcA) genes, from Slackia isoflavoniconvertens DSM22006, were cloned vector pNZ:TuR, under strong constitutive promoter (TuR). Lactococcus lactis MG1363, Lacticaseibacillus casei BL23, Lactiplantibacillus plantarum WCFS1, Limosilactobacillus fermentum INIA 584L L. 832L, harbouring pNZ:TuR.tdr.ddr, dihydrodaidzein, while strains showed also 5-hydroxy-equol dihydrogenistein. metabolization genistein combination pNZ:TuR.dzr pNZ:TuR.tdr.ddr similar results, addition correspondent strain pNZ:TuR.ifcA resulted increase production, but strains. This pattern was confirmed cow's milk supplemented incubated different constructed plasmids. Bacteria generally recognized as safe (GRAS), such species used this work, plasmids, would value for development fermented vegetal foods enriched 5-hydroxy-equol.
